Abstract: Alzheimer’s disease (AD) is characterized at histopathological level by the presence in the cerebral tissue of two specific structures: senile plaques (SP) and neurofibrillary tangles (NTFs), which are large insoluble aggregates composed respectively of beta-amyloid protein and misfolded tau protein. The former protein is mostly present in the extracellular space, while the latter is mainly an intracellular protein.
The aim of this talk is to present a mathematical approach to the modelling of the process of formation of large protein clusters in AD by means of Smoluchowski-type coagulation-diffusion equations on graphs. Originally, in (Smoluchowski 1917) a system of infinite discrete differential equations (without diffusion) was introduced for the study of rapid coagulation of aerosols. Smoluchowski’s theory was successively extended to model various physical phenomena including polymerization. The mathematical properties of the solutions of Smoluchowski-type systems have been extensively studied in standard open domains; on the other hand, the use of this kind of equations defined in the usual continuous setting in the specific context of neurodegenerative diseases is a trend well-established in the literature. The novelty of the approach presented in this talk consists in the fact that the equations for the diffusion and the agglomeration of the protein species are considered on a network, whose vertices represent functionally homogeneous brain regions and whose edges describe the connections between such regions.
Thus, in the first part of the talk we will analyse the dynamics of a polymerization process for tau protein with a possible infinite size of coagulating tau polymers, coupled with mass transfer and monomers injection, which is modelled by means of an infinite-dimensional system of Smoluchowski-type equations on a finite graph. At mathematical level, the formation of insoluble tau aggregates (NTFs) is characterized in terms of a sol-gel phase transition which is proven to occur for the proposed model at some finite time, that can be estimated in terms of the parameters of the problem. Some biological implications of the model in the context of Alzheimer’s disease will be discussed.
The second part of the talk is dedicated to the analysis of a model based on finite-dimensional Smoluchowski-type system for the dynamics of both toxic beta-amyloid and tau proteins, where the mathematical and computational novelty is the introduction of two superposed graphs: in addition to the neural network, which describes the intracellular tau protein, a second graph is needed to model the extracellular beta-amyloid protein. Some numerical simulations will be shown, where various modelling hypothesis have been tested to explore the impact of the in- teraction and the synergetic action of the two protein types that seems to be crucial for the understanding of the progression of Alzheirmer’s disease as stressed by recent biomedical literature.
